The old conventional wisdom that probably still applies, is that a player needs about a full 1.5 games to go from extreme rust to zero rust. I don't think it matters when a player gets those snaps, as long as they're at some point during the preseason.
Players that play a full load in the final preseason game(s?) often show up after Week 1 as "tired". So those last two games are definitely roster bubble time for me.
Also for me, it's not worth it to make sure every starter is completely rust free. If they're a starter and even if they have "significant" rust Week 1, it'll go down to 0 for Week 2. Actually, it's the 2nd string guys who play less that have a harder time knocking off rust in the regular season, so those are the guys I want to make sure are game ready.
